Stigmas are extra skills a character equips in the stigma slots of the character window. What fits a build depends on the slots your level has opened and on the 
Stigma Shards each stone costs to equip. This guide gives both for AION Classic 5.0 and works through a Soul Reaper build at level 65 in the skill and stigma builder.
When stigma slots open
A character has six Normal Stigma slots and six Greater Stigma slots. The Normal slots open with your level, at the same levels for every class.

Greater Stigma slots do not open with level. Each one is opened by an expansion that the game gives as a reward, so the builder asks how many you have opened, from none to six. A Greater Stigma fits only a Greater slot, and a Normal stigma only a Normal slot.
What a build costs
Equipping a stigma costs Stigma Shards, and every stone has its own price. At level 65 the Soul Reaper's Normal stigmas cost 160 or 200 shards and its Greater stigmas 160, 250 or 300. The cost of a build is the sum of its stones.

Stigma Shards.Prerequisites
Some stigmas need another stigma skill first. Spirit Light I/II needs 
Impregnable I, Impregnable II or Impregnable III, and Impregnable I/II gives Impregnable I, so equip it before Spirit Light. The builder keeps a stigma whose prerequisite is missing in its slot and marks it, so you can add the missing stigma next.
A skill fits in one slot only. Two stones that give the same skill cannot both be equipped.
Class skills at your level
Under the stigmas the builder lists the class skills at the chosen level, split into skills learned automatically and skills learned from a skill book. At level 65 the Soul Reaper's skill book skills include 
Blade Avalanche V at skill level 6 and 
Dark Pursuer V at skill level 8.
Common questions
When does the last Normal stigma slot open? At level 70. The fifth opens at level 50.
How do I open Greater Stigma slots? With expansions that the game gives as rewards, one slot at a time. Your level does not open them.
Can I equip two stigmas with the same skill? No. A skill fits in one slot only.
Does the builder keep my build? Yes. The build stays in the page link and in this browser, one build per class.
